Aveley are at home to fellow Isthmian North side Potters Bar - a clash that pits the top two sides in the division together after both clubs have made exceptional starts to the season.
Both have four wins and a defeat from their opening five matches.
East Thurrock United will be at home to Biggleswade FC from the Southern League Central Division, the same level in the non-League pyramid as Rocks.
Biggleswade sit in mid table, with a win and two draws to their credit, while Rocks languish in the lower regions of the Isthmian North after four defeats.
Grays Athltic are on the road and will travel to Edgware & Kingsbury FC of the Combined Counties League Premier Division North, who have already pulled off one cup giant-killing act by beating Southern League Division One Central side AFC Dunstable.
The winners of today's ties get £2,250, and are guaranteed at least £1,125 from the next round. Losers today walk away with £750.
Tilbury do not have a fixture this weekend as they bowed out of the cup at Waltham Abbey.
